4in1 kit: Genekam has developed a new test (4in1) to differentiate between Zika, Dengue (1-2, 3-4) and Chikungunya in one step. Such assays can be helpful to screen the donors for blood banks as the blood transfusion is one of source of infection, hence it is important to trace the such donors or blood samples, which may be positive to above said viruses. Other applications of such tests are screening of organs, cell cultures and semen donors etc. 4in1 kit can be one of important monitoring tool for mosquito borne pathogens in different clinical and non-clinical settings. Serological tests are of little use as mentioned already. Zika virus is known to cause congenital infections leading to microcephaly, which may cause of number of issues during the whole life of an indivual. Such assays can be used to screen the people in prone areas to know which virus is prevalent as it is same vector Aedes aegypti or A. albopictus, which carry all these viruses. Genekam Test is multiplex assay. Please note that there is technological disadvantage of multiplexing as some time, it is is less sensitive against simplex (if you are running each test as single test). This Genekam assay offer quick screening of samples as it will save money and time. Moreover such tests may be a great help for screening the samples from pregnant women to find whether they are positive or negative. Not only this, this test will indicate the load the viruses also. This test kit contain all components needed to run the assay. User needs to add RNA of the samples into the components and put in the realtime machine and the results can be ready within 2 hours. On one 96 well plate, one can run around 90 samples i.e. one can 90 samples tested within 2 hours.

The important part of this kit is that it differentiates between different Dengue genotypes (1-2 and 3-4) and it can be very important feature as Dengue 2 is known to cause more fatal outcome of the disease.

4in1 kit is being tested with different cross reaction panel e.g. Plasmodium, Westnile virus, JEV, HSV1 and 2, Yellow fever, HCV, HIV, HBV, CMV, Toxoplasma, Influenza, Treponema and so on. They should be negative.

This kit functions excellent as it is highly specific, sensitive and accurate. This kit has been tested with Zika virus reference strain along with strains of Dengue viruses and Chikungunya viruses. This kit detects all these strains. Virtual analysis of primers and probes show that they are highly specific for their targets and does not cross reaction with any pathogens or part of human body.

Mycobacterium tuberculosis – Double Check (Ready to use RealTime PCR-Kit)CE (FR013a)

Mycobacterium tuberculosis is the causative agent of tuberculosis (TB) in the world. It is a chronic disease, which spreads through coughing, sneezing, speaking and singing as the bacteria spread in air. There are a number of persons with latent infections of the TB around the around the world, where latently infected patient rarely experience the symptoms of the disease, but act us the source for other infections. In the active form of TB, the symptoms are loss of weight, coughing, sputum with blood, fever, chills, pain in chest, cough lasting for more than 3 weeks. TB can be a compounding factor in immunosuppressive conditions like HIV. The TB can be detected with help of tuberculin test (skin test) or interferon test and culturing the sputum or blood samples. Latent TB can not be detected properly as there is problem of cross reaction in tuberculin and interferon tests in many diseases e.g. allergies, where these tests can lead to wrong results. These test may fail to differentiate between vaccinated and non-vaccinated cases. In the literature, there are many publications, which are pointing out the drawbacks of these tests, but still these tests are being used in diagnostic regularly. Similarly weeks are needed to get the results with culturing methods. Application of serological tests should be done carefully as there may be cross reactions leading to false results. Therefore there is a need of reliable tests, which can give accurate, highly specific and sensitive results within a short period. This can be done with PCR method. Real time PCR method can give the results within a short period and it can indicate the load of bacterial in form Ct values in the samples, which can be compared between the samples in order to find the success of the therapy. PCR method is also in position to detect the latent TB. But this can be improved combining the other parameters e.g. detecting the mRNA expression of different cytokines.

TB can be cured with different kinds of therapies. The drugs available are rifampin, isoniazid, ethambutol and pyrazinamide. The course of treatment can be over the months. During the treatment, there is chance of development of drug resistance strains. The prevention of drug strains of Mycobacterium is one of biggest challenge in medicine.

The vaccination is available, therefore people should be vaccinated to reduce the TB in the world. This vaccine is based on BCG strain. The recommendation of application of this vaccine varies country to country.

Tuberculosis (TB) is one of major public health problem throughout the world. In order to overcome this disease, one has to develop rapid, sensitive and specific identifcation methods. Traditional laboratory methods such direct microscopy and culture methods have drawbacks in view of identification of Mycobacterium tuberculosis as they are not so sensitive or they need skilled manpower (who give again different results of the same probe) or need a long time. The detection of Mycobacterium tuberculosis DNA sequence might be representing a more sensitive and fast daignostic target. One needs efficient method to detect the Mycobacterium tuberculosis in sputum and moreover to monitor the drug therapy, there is need of quantitative PCR assay. At present, the laboratories are using the PCR assay, which detects only one target gene and there are a certain strains of Mycobacterium tuberculosis, which does not contain this strain, therefore they can escape this test, therefore Genekam has brough a ready to use PCR kit, which can detect one most common gene target (which is widely accepted as standard throughout the world in all laboratories carrying the identification of Mycobacterium tuberculosis) and an extra gene, which is available in certain strains without the most common gene target.

However Genekam offers the assay in two forms:
1. ready to use kit for one target gene
2. ready to use kit for detectiong 2 different target genes

target gene: one IS gene (multicopy gene) in simplex version and two highly conversed gene targets (one is IS: it is multicopy gene and an other gene) in duplex version

Types of the samples: Sputum, strains from the culture, Respiratory fluid, lung, tissue, blood etc. DNA can be isolated through commecial available kits or home made good methods. This kit need good quality of DNA.

Contents of Kit: primern, fluorogenic primers (filter FAM to used in one step PCR and filters FAM and VIC are to be used in 2 step PCR), nucleotides, buffers, molecular water, negative control, positive control and manual. Dyes used are 6-carboxy-fluorscein, YYE and 6-carboxytetramethylrhodamine.

Validations: The assay has been validated for cross reactions with related like Mycobacterium avium sp paratuberculosis, Mycobacterium avium sp avium etc and non related micro organisms like Salmonella, Legionella, Staphyloccus etc. along with the different viruses of human and animal origin. The assy is 98% sensitive in most common used IS gene against the microscopic method.

Genekam Stem cell Isolator (sterile) 100ml (STEM13)

Isolation Solution: Genekam Stem Cell Isolater*

Genekam Stem cells isolater from Umbilical blood samples: Genekam Biotechnology AG has developed a simple, economical method to isolate stem cells from umblical cord. You just add the solution into umblical blood and centrifuge it. You can isolate the stem cells from small volumes of fluid also e.g. 100 µl (provided they contain the stem cells) as well as larger size samples. Now you can isolate the stem cells with different methods as well as you can do further analysis. This method is far better than density gradient method to isolate the stem cells.
Genekam Stem cell (SC) isolator from umbilical cord blood is ready to use sterile solution for isolating stem cells containing populations (SCP) quickly from blood, buffy coats, bone marrow aspirates etc of different mammals like human, rat, primates, cattle, sheep, goat, pig, horse, camel, dog, cat etc.

This kit contains followings:

SC isolator
Washing buffer
Here are a few examples of application of SC isolater:

A. Isolation of SCP from umbilical cord blood:

Add 900 µl of Genekam SC isolater to each tube containing up to 100 µl of umbilical cord blood in one sterile tube as this is ration (1:10) of solution to the sample e.g. if you want to isolate cells from 4 ml blood of and add 36 ml of SC isolator. You have to decide howmuch blood sample do you want to use to isolate SCP as you will get a lot of SCP against the conventional method of density gradient centrifugation.
Warning: Kindly work with smallest volume of sample to gain some experience with this kit instead of destroying whole sample for first time.
Gently vortex each tube immediately after adding the solution. Keep at room temperature, protected from light, for 7-10 minutes.
Centrifuge 1600 rpm for 5 minutes. Aspirate or discard supernatant without disturbing pellet.
Resuspend the pellet in washing buffer and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the cell number and go for your isolation of stem cells e.g. cell culturing or magnetic beads.
Stem cells can be isolated through pouring the isolated SCP (1x 106/cm2 suspended in DMEM with 10-15% FCS or other media; it is recommended to use FCS precoated plates as they may increase the chances to get the stem cells) into cell culture plates. Keep them at 37°C and 5% for 24-72 hours. After that discard the supernant as the stem cells will be adherent on plastic as adherent fibroblastiod cells, they can be grown and subcultured. Subconfluence of these cells can be isolated with trypsin. Usually cells at the second passage and thereafter can be replated 3.5 to 5 x 103/cm2.

B. Isolation of SCP from human bone marrow aspirate:

Add 400 µl of Genekam SC isolater to each tube containing up to 200 µl of human bone marrow in one sterile tube as this is ration of solution to the sample (1:3 ration) e.g. if you want to isolate SCP from 4 ml of bone marrow aspirate and add 8 ml of SCP isolator. You have to decide about the volume of bone marrow aspirate you want to use to isolate the cells because you will get a lot of cells against the conventional method of density gradient centrifugation.
Warning: If you donot have any experience with this kit, kindly use smallest volume of sample to have experience with this kit before you destroyed the whole sample. You may have to adjust ration e.g. you have still SCP containing some erythrocytes at the end, you have to use more SC isolator (1 volume sample and 4 volume SC isolator e.g. 200 µl sample and 600 µl of SC isolator) or wash one time more. Usually the above said protocoal in point 1 is good enough to isoate.<

Osteogenic differentiation media (10X) (STEM05)

Kit contents: STEM05 Genekam osteogenic differentition media (10X) 10ml (store it at -20°C)
Prepare the media while adding 10 ml of Genekam osteogenic differentiationmedia into 90ml of DMEM containing 10 % FCS + 1% SP + 1% Glutamine or in your culturing media. Store it at 4°C. You can prepare less media, but the ratio should be 1:10.

After you have stem cells e.g. MSC. To isolate MSC cells, one can use Genekam Stem cell isolator kit. After you get 60-85% confluence during culturing of stem cells, please remove the cell using trypsin or Genekam cell remover. and wash the cells to make free from trypsin while Centifuging 1400 rmp for 5 minutes and suspend the pellet now to make the cell count using trypan blue.
Now add the cells 3-5 x 103 / cm2 on a plate e.g. 9 or 12 wells in osteogenic differentiation media (containing DMEM + 10 % FCS or other serum source + Genekam osteogenicc differentiation media) for two weeks while feeding the cells 3-5 days with this media for 3 weeks.
After two weeks, you can stain with cells with Alizarin Red S to see the bone formation.

Mycoplasma pneumoniae is a respiratory pathogen, which causes around 10-20% of all community acquired pneumoniae in human beings.It is very difficult to make the differentiate diagnosis between the patients suffering from Chlamydia pneumoniae and Mycoplasma pneumoniae, therefore there is a need of laboratory diagnosis. This laboratory diagnosis can be done through culture methods (which takes a longtime as Mycoplasma goes slowly and needs special media), serological methods like ELISA as well as FAT, which are prone to cross reactions, therefore the PCR is merging as a goldstandard for detection of micro organisms in medicine as this is highly sensitive and specific also. Moreover PCR needs a little material to conduct the test.

Target gene: highly conserved region of repetitive gene of Mycoplasma pneumoniae. Around 8% genome of Mycoplasma pneumoniae is made of repetitive DNA elements.

Kinds of samples: Throat swabs, respiratory system swabs, colonies grown on cultures, reproductive tract swabs, blood samples, tissue samples and other body fluid samples. This kit needs isolated DNA, which can be isolated using commercial available kits or home made methods.

Components of the kit: polymerase, forward and backward primers, nucleotides, buffers, PCR master mix, loading dye, gene ruler (100 bp), positive control, negative control as well as manual.

Validations: It has been tested for cross reactions with related and non related micror organisms like Mycoplasma genitalium, Chlaymdia trachomatis, Chlaymdia pneumoniae, Mycoplasma hominis, Ureaplasma urealyticum, Legionella pneumoniae, Styphococcus aereus, Bordetella pertusis, CMV, RSV and many more. It is tested for serial dilutions.

GENEKAM Magnetic separation of CD4 positive cells (usually Human T-cells) (STEM23)

Magnetic cell separation from blood: In blood, there are many cells, which can be isolated to conduct various experiments like developing new kinds of assays and cell therapies. Separation of cells can be also done through magnetic field. This method is quite simple and can be conducted in laboratories without the help of complex and expensive instruments. Therefore Genekam Biotechnology AG has developed a number of solutions to do. These are different markers being expressed on the cells, which can be used as source of isolation in order to get pure population. After the isolation, one should do various experiments like microscopic and flowcytometry examination to be sure that the quality of cells is sufficient good to go ahead with further work. The offered solutions are sterile, hence isolated cells under sterile conditions can be used for cell cultures. These isolated cells can be used for molecular and immunological analysis. This kit can be used to isolate cells from as little as 1 ml of blood.

Isolation kit includes: magnetic beads, Fc-Receptor blocker, enrichment solution of white blood cells
The kit contains also components for enrichment of white blood cells, hence one can save time and money for density gradient method. However one can also use density gradient method, if it is essential to isolate the cells.

These are different markers being expressed on the cells, which can be used as source of isolation in order to get pure population. Human circulating tumour cells (CTC) in blood are an important indicator of the progression of a cancer and in some cases, these may be an indication of so called sleeping tumour cells i.e. it does mean that there is an active disease. Therefore PCR tests are being conducted in many laboratories around the world for presence of markers of such blood cells. As the number of such cell is very low, therefore there is a need of enrichment of these cells before doing RNA isolation. Genekam Biotechnology AG has developed this enrichment step and this can be combined with mini-column method or other method. Genekam offers this enrichment step as sterilised product because some users may want to grow enriched cells in order to do other studies including nucleic cells isolation e.g. tumour resistance (if possible!), but that one can go directly to RNA isolation step after enrichment even under unsterilised environment in order to run the tumour marker PCRassay. There is only 1 circulating tumour cell in one million of mononuclear cells, therefore enrichment is very important step during the conducting PCR for human cancer markers from blood. Peripheral blood is one of easiest way to access against the biopsy of lymphnodes or bone marrow, hence it is an excellent idea, if one can develop more and more assays based on CTC.

Important feature of enrichment step is that one can do the isolation of circulating tumour cells directly from blood 1ml or 3ml and there is no need of using density gradient method.

World Innovation and Hope: First therapeutic molecule candidate against Zika viruses

Zika viruses are known to cause fears as they infect pregnant women leading to disorders and new born with microencephaly (child with smaller head) with whole life handicap and missing a number of vital functions e.g. eyes, limbs, speech etc. Till today diagnostic are only the choice to detect and prevent the Zika viruses. There are claims that vaccination candidates are being developed and tested. But there is no claim in the world that any company has got the potential therapeutic molecule for Zika viruses. The experiments show that Genekam Zika-molecule are inhibiting the Zika virus infection and likely to be non toxic. But still there is a need of more studies till these molecules go for trial on human beings.
