Advancing immune cell analysis with micro-robots

Researchers at the University of Chicago have created tiny, spinning micro-robots, dubbed “hexapods,” designed to bind to immune cells and investigate their behavior. Each hexapod is equipped with six arms containing molecules potentially recognized as foreign by the immune system, enabling scientists to explore interactions with immune cells and enhance immunotherapy development.

Jun Huang, co-senior author of the study, highlights the significance of this innovation in elucidating molecular interactions within the immune system. By utilizing inorganic materials, researchers delve into previously unexplored realms of immune cell manipulation, offering new avenues for scientific exploration.

T cells play a crucial role in identifying foreign pathogens processed by dendritic cells. However, pinpointing the specific T cell recognizing a pathogen among trillions presents a significant challenge. Traditional methods rely on T cell receptor binding, which may not always provoke a robust immune response. The hexapod system provides a more accurate approach, mimicking the role of physical force in dendritic cell-T cell interactions and facilitating precise immune cell targeting.

To overcome existing limitations, researchers designed robotic mimics for dendritic cells, featuring a central magnetic core and six arms for antigen attachment. These hexapods effectively identified the correct T cell receptor, even amidst numerous other cells, showcasing remarkable accuracy and efficiency.

The study also delved into analyzing the immune responses of T cells bound to hexapods. By examining immune activity and force exertion, researchers gained insights into the dynamics of immune cell interactions. The findings highlight the potential of hexapods in elucidating immune response mechanisms and advancing immunotherapy strategies.

Looking ahead, researchers aim to extend hexapod applications to various antigens, including those associated with human cancers and pathogens. This innovative approach holds promise for identifying T cells with robust reactions to specific antigens, paving the way for tailored immunotherapy solutions.
