Genekam endotoxin removal Kit (to remove endotoxin): There are many applications, where endotoxins are causing serious problems. Many groups from institutes, universities and biotech industry are working on recombinant proteins expressed in bacteria like E. coli and othe organisms, hence the isolated proteins from much cultures contain a lot of lipopolysaccharides (LPS) so called endotoxins, which can alter the results of isolated proteins during their applications, therefore there is an urgent need to remove them before applying these proteins as therapeutics, diagnostics reagents (medical devices), antibodies (humanised / other origin), vaccines etc. and it is highly recommended to remove them from recombinant proteins in order to start in vitro, preclinical as well as clinical studies. The presence of endotoxins in injectable compounds may lead to sepsis (septic shock!) resulting in the failure of a good compound. Endotoxins may lead to serious problems during the cell cultures. The source of endotoxin may be water, components as well as sera to be used during the culturing or fermentation process. There are different methods available to remove them so e.g. ultracentrifugation, ultrafiltration, chemical treatment etc. None of the methods is going to remove the endotoxin fully, hence a combination of these methods may be alternative. Chemical treatment of isolated protein may leave traces of the chemical leading to side effects during the therapy or development of therapies. Genekam has developed magnetic beads to remove them from isolated proteins. The method is very simple as one can add magnetic beads to the solution, keeps it for 30 minutes and remove the beads (on which the endoxins bind) with a magnet or magnetic field. Genekam beads are designed in such a way that they will remove the maximum endotoxins in the first attempt. However it may be possible that one has to repeat it to remove the endotoxins depending upon the grade of contamination. Genekam beads can be used to remove the LPS from cell cultures, liquids (including water) etc. Even these beads can be used to remove endotoxins from nucleic acid solutions. At present, Genekam offers to remove the endotoxins from small volume samples. If the user has a need to remove endotoxins from large scale samples, Genekam offer here solutions for large scale user.There are many companies around the world, which are producing recombinant proteins or other products for cell therapies, advanced tissue therapies etc. One should test the products for the presence of endotoxins before applying them for different uses. Even the cell culture media should be tested to meet the lowest possible endotoxin concentrations. Technology of Genekam is of great help. Genekam endotoxin removal kit is a sterile product, hence it can be used in sterile applications like cell cultures and sterile proteins (to be used for in vivo research applications). One can regenerate used magnetic beads to be used again. There are many products for which there are legal requirements in EU or USA from FDA, which must be met. During the fermentation process, one uses a number of bioreactors, hence the surface of reactors may be contaminated with biofilms containing endotoxins, we think that one can use Genekam magnetic beads to reduce the load of endotoxins after chemical or mechanical treatment of these reactors. Even Genekam magnetic beads may be the part of the mechanical treatment of bioreactors. These beads can be used to remove with magnetic field or with water from bioreactors. Glasswares may be one of major source of the endotoxins contamination as endotoxins bind on glass surface very strongly. Heating does not remove the endotoxin fully, therefore Genekam magnetic beads should tried to remove them too. Genekam Erythrocyte removal kit

A. During isolation of cells like mononuclear cells from different sources, many times some of erythrocytes remain in the pellet as unwanted effect. These small number of erythrocytes are problem as there is hardly any method to remove them quickly from bone marrow, tissue as well as pathological lesion. Even to isolate the erythrocytes from the solutions containing circulating tumour cell populations from blood, bone marrow as well as lymphnode biopsies. There are groups working on CAR T-cell therapies, removal of all erythrocytes should be needed to have pure population of mononuclear cells. Similarly during stem cells isolation from umbilical cord blood, erythrocytes may be needed to be removed. There are many more examples e.g. from isolation of particular cell population from umbilical cord wall, the erythrocytes needed to be removed. These are examples, where these erythrocytes are needed to be removed. There are also many applications, where user needs intact erythrocytes to conduct the microscopic analysis as in case of erythrocyte invading pathogens like plasmodium, anaplasma, babesia etc. Genekam has developed the method to isolate these intact erythrocytes with magnetic field, hence one can get erythrocyte free pellet to do further analysis. There will be hardly any lysis of erythrocytes because they will be removed as intact cell population. This kit is very simple to be used (just add Genekam solution, keep for a few minutes and remove the erythrocytes with magnetic field).

During isolation of cells e.g. white blood cells populations like mononuclear cells from blood, spleen, umbilical blood cord, bone marrow etc, there are often a problem that erythrocytes are mixing in the population. Content:

Genekam Eryhtrocyte removal solution (50X)

Equipment needed:

Magnetic rack

PBS optional

Pipettor

Procedure: 1. Prepare the ready to use solution in washing PBS without any serum or other additive. ( 2ml of Genekam Erythrocyte removal in 98 ml PBS to get ready to use solution because it is 50X): Now make the pellet of isolated cells containing erythrocytes and suspend this pellet in 4-6 ml Genekam erythrocyte removal solution. Keep it at 4 °C for 20 to 30 minutes. Now put the tube in magnetic rack and keep it for 3 minutes. (one will observe that erythrocytes are attractive to the wall through magnetic field). Remove the supernatant from the tube containing the cells free from erythrocytes with pipettor carefully. The pellet or spot containing erythrocytes can be seen as a spot on the wall of the tube because the magnet has attracted the treated erythrocytes. There should be no lysis of erythrocytes!

Hint: If user wants to conduct microscope analysis of erythrocytes containing pathogens. The erythrocytes can be collected as separate population. The erythrocytes can be placed on the microscopic slide for further analysis.

2. Now centrifuge the pellet 2000 rpm for 10 minutes to get the pellet containing cells without erythrocytes.

3. Wash the pellet with PBS and centrifuge it as step 2. This pellet can be used for different applications.

B. Genekam MNC isolator can be used with above said method to isolate mononuclear cells in large quantity directly from blood. These MNC population can be used for to develop CAR T-cell therapies, other theapies as well as cell culturing.

Genekam DNA isolation kit

Genekam DNA isolation kit (DNA isolation cost as little as 10 cents: perhaps the cheapest isolation kit of the world): It can be used to isolate the DNA from blood samples, bacterial colonies, plasmids, blood stained cloths, blood stained wood, seminal fluid, sperms as well as also buccal swab for human, animals and birds. It can be used with the tissue also. This can be used with mouse tail or ear tips also for mouse genotyping . There are many pathological samples, where wax may create problem. We think that pathological laboratories should try our this method (remove the wax with Xylene and put a small piece of tissue in our kit to get isolated DNA. Please note that this kit is made in our own laboratory. 

Blood samples on filter paper or normal paper: One can put one or two drops of blood on normal paper or filter paper. After that one can dry this and inactivate this with ethanol. Now these samples can be sent in an envelope throughout the world. Genekam DNA isolation kit can be used to isolate the DNA from such blood samples. These samples can be stored at room temperature for months or perhaps for years. As you need only 15-30 seconds to isolate the DNA from one sample i.e. one can isolate 100 samples within one hours, therefore this can be the method of choice for many laboratories and one can save the expensive automatic instruments. This method is being used in our laboratory for last 13 years to do the PCR analysis. In case one wants to store the sample. One can divide the paper in two parts. One part can be processed and other part can be stored for long-time. Many laboratories around the world are using it now. 

Advantages:

Usually many kits on the market fail to do the isolation of DNA from blood (without removing the red blood cells). In our case you can isolate the DNA directly from blood. Similarly many kits fail with sperms. Various samples like blood samples, oral swabs, milk, meat / tissue can be used.
This kit is made on the method, which is used for last 13 years in our laboratory to isolate the DNA from blood samples of birds, dogs and humans.
We found that our kit is 100 times more sensitive than kit of most of expensive and most used manufacturer in the world (this is done with house keeping gene for human as well as birds). This is the result of our internal studies.

Hence the laboratory can request their clients to send 3-4 drops on filter paper in simple envelope for detection of mutations like genetic disorders as well as for other purposes. In this way, one can send 20-100 probes in simple envelop throughout the world. Precaution: the blood probes on filter paper can inactivated with ethanol (94% or 99%) so that there should be no risk for environment i.e. inactivation of infectious agents. Genekam DNA isolation kit is environment friendly too as there will be no waste and no need of extra plastic ware.
As there is no loss of DNA during the isolation process, therefore it may be possible that you have highly concentrated DNA, hence use 1 or 2 ul in your PCR reactions. In some cases, you may need 0.5 ul. The concentration of isolated DNA depends on your sample.

Genekam Quick MNC Isolator (sterile) 25ml

Isolation Solution: Genekam Quick MNC Isolator*

Genekam Biotechnology AG has developed a new method to isolate the mononuclear cells (MNC) from different sources like bone marrow aspirates, buffy coats, blood, umbilical cord blood and other sources. Usually one uses density gradient centrifugation, which is a time consuming and need a lot of precautions during the pipetting of fluids on density gradient. Making a mistake means that you are going to lose the whole samples. Density gradient methods are not good for isolation of MNC from the sources, where you have less than one ml volume and more you get only a part of MNC as you cannot take out the cells from white ring. Genekam Biotechnology AG has developed one method, where you can isolate all cells during your isolation available in sample within 20 minutes. Not only this method is faster, but it is also economical and let you isolate the MNC from as little as 25µl of samples along with the larger size samples. This is a sterile product.

Genekam Quick MNC isolator is ready to use sterile solution for isolating the mononuclear cells (MNC) quickly from blood, buffy coats, bone marrow aspirates, spleen etc of different mammals like human, rat, primates, cattle, sheep, goat, pig, horse, camel, dog, cat etc.

Here are a few examples of application of Quick MNC isolater:

A. Isolation of MNC from human peripheral blood or buffy coat:

Add 900µl of Genekam Quick MNC isolater to each tube containing up to 100 µl of whole blood or buffy coat in one sterile tube as this is ration (1:5) of solution to the sample e.g. if you want to isolate MNC from 2ml of buffy coat and add 8 ml of Quick MNC isolator. You have to decide howmuch blood or buffy coat sample do you want to use to isolate the MNC as you will get a lot of MNC against the conventional method of density gradient centrifugation.
Warning: Kindly work with smallest volume of sample to gain some experience with this kit instead of destroying whole sample for first time.
Gently vortex each tube immediately after adding the solution. Keep at room temperature, protected from light, for 7-10 minutes.
Centrifuge 1600 rpm for 5 minutes. Aspirate or discard supernatant without disturbing pellet.
Resuspend the pellet in the appropriate buffer (usually it is PBS in most of applications) and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the number of MNC and go for your analysis e.g. cell culturing.
B. Isolation of MNC of human bone marrow aspirate:

Add 400µl of Genekam Quick MNC isolater to each tube containing up to 200µl of human bone marrow in one sterile tube as this is ration of solution to the sample (1:3 ration) e.g. if you want to isolate MNC from 2 ml of bone marrow aspirate and add 4 ml of Quick MNC isolator. You have to decide about the volume of bone marrow you want to use to isolate the MNC because you will get a lot of MNC against the conventional method of density gradient centrifugation.
Warning: If you do not have any experience with this kit, kindly use smallest volume of sample to have experience with this kit before you destroyed the whole sample as you may have to adjust ration e.g. you have still erythrocytes at the end, you have to use more MNC isolator (1 volume sample and 4 volume MNC isolator e.g. 200 µl sample and 600 µl of MNC isolator) or wash one time more. Usually the above said protocol is good enough to isolate MNC.
Gently vortex each tube immediately after adding the MNC isolator solution. Keep at room temperature, protected from light, for 7-10 minutes.
Centrifuge 1600 rpm for 5 minutes. Aspirate or discard supernatant without disturbing pellet.
Resuspend the pellet in the appropriate buffer (usually it is PBS in most of applications) and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the number of MNC and go for your analysis e.g. cell culturing.

C. Isolation of MSC from of mouse spleen:

Prepare a single cell suspension after harvesting the mouse spleen.
Pellet the cells by centrifugation (1600 rpm); discard the supernatant.
Now suspend the pellet in 5 ml of MNC isolator isolation.
Incubate at room temperature for 7 minutes with occasional shaking.
Add to MSC isolator solution with spleen cell with 20-30 ml of PBS buffer.
Spin the cells (1600 rpm) for 5-7 minutes and discard the supernatant.
Resuspend the pellet in the appropriate buffer (usually it is PBS in most of applications) and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the number of MNC and go for your analysis e.g. cell culturing.

Genekam Stem cells isolator from Umbilical blood samples: Genekam Biotechnology AG has developed a simple, economical method to isolate stem cells from umblical cord. You just add the solution into umblical blood and centrifuge it. You can isolate the stem cells from small volumes of fluid also e.g. 100 µl (provided they contain the stem cells) as well as larger size samples. Resuspend the pellet in washing buffer and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the cell number and go for your isolation of stem cells e.g. cell culturing or magnetic beads.
Stem cells can be isolated through pouring the isolated SCP (1x 106/cm2 suspended in DMEM with 10-15% FCS or other media; it is recommended to use FCS precoated plates as they may increase the chances to get the stem cells) into cell culture plates. Keep them at 37°C and 5% for 24-72 hours. After that discard the supernant as the stem cells will be adherent on plastic as adherent fibroblastiod cells, they can be grown and subcultured. Subconfluence of these cells can be isolated with trypsin. Usually cells at the second passage and thereafter can be replated 3.5 to 5 x 103/cm2.

B. Isolation of SCP from human bone marrow aspirate:

Add 400 µl of Genekam SC isolater to each tube containing up to 200 µl of human bone marrow in one sterile tube as this is ration of solution to the sample (1:3 ration) e.g. if you want to isolate SCP from 4 ml of bone marrow aspirate and add 8 ml of SCP isolator. You have to decide about the volume of bone marrow aspirate you want to use to isolate the cells because you will get a lot of cells against the conventional method of density gradient centrifugation.
Warning: If you donot have any experience with this kit, kindly use smallest volume of sample to have experience with this kit before you destroyed the whole sample. You may have to adjust ration e.g. you have still SCP containing some erythrocytes at the end, you have to use more SC isolator (1 volume sample and 4 volume SC isolator e.g. 200 µl sample and 600 µl of SC isolator) or wash one time more. Usually the above said protocoal in point 1 is good enough to isoate.<
Gently vortex each tube immediately after adding solution. Keep at room temperature, protected from light, for 7-10 minutes.
Centrifuge 1600 rpm for 5 minutes. Aspirate or discard supernatant without disturbing pellet.
Resuspend the pellet in washing buffer and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.

DNA needed per square centimeter space is around 25 to 250 ng. This can help to calculate the total requirement for experiments.

Human circulating tumour cells (CTC) in blood are an important indicator of the progression of a cancer and in some cases, these may be an indication of so called sleeping tumour cells i.e. it does mean that there is an active disease. Therefore PCR tests are being conducted in many laboratories around the world for presence of markers of such blood cells. As the number of such cell is very low, therefore there is a need of enrichment of these cells before doing RNA isolation. Genekam Biotechnology AG has developed this enrichment step and this can be combined with mini-column method or other method. Genekam offers this enrichment step as sterilised product because some users may want to grow enriched cells in order to do other studies including nucleic cells isolation e.g. tumour resistance (if possible!), but that one can go directly to RNA isolation step after enrichment even under unsterilised environment in order to run the tumour marker PCRassay. There is only 1 circulating tumour cell in one million of mononuclear cells, therefore enrichment is very important step during the conducting PCR for human cancer markers from blood. Peripheral blood is one of easiest way to access against the biopsy of lymphnodes or bone marrow, hence it is an excellent idea, if one can develop more and more assays based on CTC.

Important feature of enrichment step is that one can do the isolation of circulating tumour cells directly from blood 1ml or 3ml and there is no need of using density gradient method.
