AKLIDES® - 1st Fully automated digital IFA System

The AKLIDES® technology was the first automated system for the immunofluorescence (IF) standardization in routine laboratories for autoimmunity. Today AKLIDES® is also applicable in the field of cell damage and microbiology.

Main features of the automated system are:
Analysis
Analysis of fluorescence patterns and quantification of fluorescence intensities is provided by AKLIDES®. The system offers a standardized positive-negative decision and pattern recognition in less than 30 seconds per well. Assays for ANA, ANCA, nDNA, tissues and CytoBead® are available with fully automated analysis.

Image Storage
AKLIDES® automatically stores the fluorescence images and defined patterns of each sample. This allows digital archiving over years and searching for results in an internal database.

Intensity Calibration
The AKLIDES® calibration slides are characterized by stable fluorescence for years. Yearly calibration leads to an even better standardization.

Titer Detection
Due to the AKLIDES® titer detection algorithm titration of patient samples for the cell-based assays is no longer needed. The AKLIDES® Doctor software allows access to the AKLIDES® analysis data from every work space. With the appropriate network structure, working via internet is also possible.

Export Options

Anti-Phospholipid 10 Dot - Profiling immunoassay for the determination antibodies to 10 phospholipids and serum proteins

Laboratory diagnosis criteria of anti-phospholipid antibody syndrome (APS or APLS) includes the detection of lupus anticoagulant as well as antibodies against cardiolipin and/or β2-glycoprotein I, and remain challenging since APS antibodies can be associated with several infections.

The immunoassay Anti-Phospholipid 10 Dot offers for the first time the possibility to profile APS antibodies detection in serum, along with the criteria antigens cardiolipin and β2‑glycoprotein I, also other non-criteria 6 phospholipids and 1 additional serum protein.

In contrast to commercially available ELISA Kits, the Anti-Phospholipid 10 Dot due to its optimal orientation of the antigens allow the correct binding of patient plasma protein cofactors, e.g. β 2 glycoprotein-I (β2 GP-I), leading to exposure of the specific epitope for the APS related antibodies.

The large number of antigens and the optimal exposure of the epitope contribute to a high specificity in diagnosis of APS, better subtyping and easier differential diagnosis of APS patients.  

The test gives reliable results in serum in less than 2h and can be run easily manually or completely automated using a DotDiver2.0.

APS, also known as Hughes syndrome, is an autoimmune disease in which antiphospholipid antibodies react against proteins that bind to anionic phospholipids on plasma membranes. The clinical symptoms include arterial and venous thrombosis, thrombocytopenia and recurrent fetal loss; stillbirth, preterm delivery, and severe preeclampsia.

APS can be primary, in which case the autoimmunity occurs in the absence of other related diseases; or secondary APS, which occurs in combination with other autoimmune diseases, such as systemic lupus erythematosus (SLE). On rare occasions, APS may lead to rapid organ failure due to generalized thrombosis, termed "catastrophic antiphospholipid syndrome" (CAPS or Asherson syndrome) which is associated with a high risk of death.

In the investigations of a peripheral neuropathy with a possible immunological background detection of antibodies against gangliosides is considered very important for the diagnosis, but also provides therapeutically relevant information regarding the subtype of neuropathic disease (i.e Guillain-Barré syndrome, Miller-Fisher syndrome and others).

Gangliosides are acidic glycolipids, comprised of a lipid (ceramide), an oligosaccharide chain and sialic acid. Inflammatory neuropathies of the peripheral nervous system are characterized by numerous clinical symptoms, which can range from mild fatigue and uncharacteristic discomfort to neuromuscular disorders and functional deficiencies, like respiratory failure and cardiac arrest.

Autoantibodies against gangliosides occur in autoimmune diseases of the peripheral nervous system. Gangliosides are components of cell membranes and are particularly found in the central and peripheral nervous system. These antibodies directly cause an inflammation or demyelinization of the myelin sheaths, therapy of autoimmune neuropathies is therefore based on immunoregulation.

Non-specific inflammatory bowel disease including Crohn’s disease (CD) (Enteritis regionalis) and ulcerative colitis (UC) are characterized by chronic-remitting inflammatory processes of the intestine.

GA Generic Assays Anti-GP2 ELISA determine antibodies to the major antigen of the exocrine pancreas, the membrane-bound pancreatic Glycoprotein 2 (GP2). Antibodies against GP2, in combination with the detection of autoantibodies to Saccharomyces cerevisiae (ASCA), which is highly prevalent (70 %) in patients with CD and atypical antineutrophil cytoplasmatic antigens (aANCA), mainly found in patients with UC, could be used as a highly specific serological marker for differential diagnosis of CD to UC.

GA Generic Assays Anti-GP2 ELISA is used for the quantitative determination of IgA and IgG antibodies against glycoprotein 2 (GP2) in human serum.

Assay Basics:
SELco® TRAb human 1-step is a competitive radio receptor assay (RRA). During the incubation step the TSH receptor autoantibodies (TRAb) of the patient samples or calibrators compete with iodinated monoclonal antibodies (Tracer) to TSH receptor for the immobilized TSH receptor on the solid phase of coated tubes.
The level of the measuring signal is inversely proportional to the TRAb concentration in the tested samples and can be read off from a calibrator curve with known TRAb concentrations.

Scientific Background
Graves' disease is an autoimmune disease of the thyroid gland. In this disease the body's own defense cells form antibodies against receptors of the thyroid cells, resulting to increasinged production of thyroid hormones, known as hyperthyroidism. The consequences are numerous disorders, such as tachycardia or cardiac arrhythmia, weight loss amidst ravenous appetite, restlessness and nervousness.

The determination of the TSH receptor antibodies (TRAb) is a major step in the diagnosis of Graves' disease.
